POSTECH Develops RNA Gene Circuits for Self-Decision-Making by Cells… “One Step Closer to Realizing Living Computers”

A new technology has been developed in which ribosomes, which are responsible for producing proteins within cells, now also perform the function of a “switch” controlling gene expression. This is expected to enable the implementation of advanced gene circuits where cells can independently analyze and interpret various signals.

A research team led by Professor Jong-Min Kim of the Department of Biological Sciences at POSTECH (Pohang University of Science and Technology) announced the development of “RATEX,” an RNA-based gene circuit platform capable of simultaneously processing multiple signals within a cell. The research involved Dr. Kang Han-sol and integrated graduate researchers Hyun-Seop Ko and Chae-Ri Kim, and the findings were published in the international academic journal Angewandte Chemie.

Gene expression in living organisms consists of the transcription process, which transfers DNA information to RNA, and the translation process, which synthesizes proteins based on RNA. Existing RNA-based gene circuits have primarily focused on recognizing and processing signals during the translation stage, which has limited capabilities for complex information processing.

To address this issue, the research team focused on ribosomes, the devices responsible for protein production. The researchers designed ribosomes to stop at designated locations on genes under specific conditions, utilizing this process itself as a signal to determine whether or not to express the gene.

The research team named this technology "Translation-Transcriptional Switching (TTC)." By directly linking the information processing performed during translation to the transcriptional regulation stage, they implemented a new platform capable of performing more complex computations than existing gene circuits.

In particular, this technology is attracting attention for its ability to design more scalable gene circuits by combining previously developed RNA logic switches.

Experimental results confirmed a gene expression regulation performance up to 1,492 times higher, and the team also succeeded in implementing a multi-logic circuit capable of simultaneously analyzing up to six RNA signals.

Furthermore, they developed a hybrid logic gate that recognizes not only RNA signals but also various metabolites such as amino acids and vitamins. This suggests the possibility for cells to process different types of information in an integrated manner.

Moreover, the research team combined this technology with a CRISPR gene control system to successfully alter cell shape or rearrange internal cellular structures only when specific conditions were met. This is considered a case demonstrating that cells can be precisely programmed to suit desired functions.

The research team anticipates that this achievement will be widely utilized in the fields of biopharmaceuticals and environmental science in the future. They explain that there is potential for application in the development of next-generation smart therapeutics that generate therapeutic substances by detecting signals found only in cancer cells, or biosensors that react only to specific pollutants.

Professor Jong-Min Kim stated, "The core of this research lies in directly linking the information processing that occurs during the translation stage with transcriptional control," adding, "We have presented a new gene circuit design strategy capable of integrally processing different signals, such as RNA and metabolites, within a single RNA design."

Meanwhile, this research was conducted with support from the Basic Research Program of the Ministry of Science and ICT and the National Research Foundation of Korea, as well as the POSTECH Institute for Basic Science Support Program, the National Research Facilities and Equipment Center of the Korea Basic Science Institute, the Food Tech R&D Hub Center of Gyeongbuk Technopark, the High Value-Added Food Technology Development Program of the Ministry of Agriculture, Food and Rural Affairs, the Health and Medical Technology R&D Program of the Ministry of Health and Welfare, and the Basic Research Program of the National Research Foundation of Korea under the Ministry of Education.
